Overview | A complete mammalian proteome extraction kit (C-PEK) designed to extract virtually all proteins from both mammalian tissues and culture cells in a simple two step process within a microcentrifuge tube to minimize protein loss. The extraction results in a total protein sample that is a clarified and non-viscous protein solution showing higher resolution and an increased total number of spots in 2-D gel electrophoresis. Sample size: 1x107 - 1x108 cells or 50-100 mg tissue. |

Materials Required but Not Delivered | ? High purity water, conductivity<5 μS (e.g., Merck) ? Cell culture equipment, media etc. for cell growth (e. g., RPMI, DMEM). ? Micropipettes and tips, 10 μl, 200 μl and 1 ml size (e.g., Eppendorf, Gilson or equivalent) ? Cooled centrifuge and rotor for 50 ml tube size (Eppendorf, Heraeus, Nalgen, etc.) ? Cooled microcentrifuge and rotor up to 25,000 x g for 2 ml tube size (Eppendorf, Heraeus, Nalgen etc.) ? Thermomixer or rolling facility (e.g., Eppendorf) ? Optional: Ultracentrifuge and rotor up to 75,000 x g for 2 ml tube size (e.g. Beckman, Sorvall etc.) ? For tissue extraction: Mortar and pestle made from porslin, small spatula. These instruments must be cooled to -70°C or colder prior to be used in the C-PEK procedure! |

Storage Conditions | ? Wash Buffer The Wash Buffer is provided ready-to-use. The buffer should be stored at 4°C. ? Resuspension Buffer The Resuspension Buffer is provided ready-to-use. It can be stored at 4°C up to 6 month. For prolonged storage freeze the buffer in convenient aliquots at -20°C. Avoid repeated freezing and thawing! ? Extraction Reagent The Extraction Reagent is supplied as dry powder and can be stored unopened at 4°C. The Extraction Reagent should be reconstituted just prior to use as described below. After reconstitution, it can be stored at 4°C up to 6 month. For prolonged storage freeze the buffers in convenient aliquots at -20°C. Avoid repeated freezing and thawing! ? Reducing Agent The Reducing Agent is supplied as dry powder. It can be stored unopened at 4°C. The Reducing Agent should be reconstituted just prior to use as described below. After reconstitution, aliquot the unused material in 60 μl volumes and freeze at -20°C for further use. Avoid repeated freezing and thawing! ? Benzonase? Benzonase? should be stored at 4°C. |
